I had this issue occur on a Windows 2008 system.  These are the steps I took to resolve:
0. First, make sure you have the appropriate permissions to the file (if you are an administrator, then you likely have the permissions needed).  This won't work if the security tab is not present - so continue on.
1. Try to rename file, if fail, go to next step
2. Right-click on a Command Prompt shortcut, choose Open As Admin, then try to rename the file using the "ren" command.  If fails, go to next step.
3. If this is still an issue, the file is either locked OR corrupt.  Since I am an optimist, I choose to check to see if the file is locked.  I have used several programs in the past for this (such as WhoLockMe) and then try to end the offending program that has the file locked.
4. If this doesn't work, then the file system could be corrupt, so using that admin launched Command Prompt window, change the prompt to the drive letter you wish to fix and type: chkdsk /f
5. If all of this fails, hopefully you can boot the system using something like BartPE or a Linux Live CD, which you can then use to hopefully delete the file (just note that using these portable OSes can be a little painful to get working, depending on the hardware in the system).

I have a log file that has grown very large (over 9GB).  The application has since started a new file but I'd like to delete the old file to recover disk space.  However when I try to perform any operations on the file Delete, Rename, dir>file.log they all return Access Denied.  I'm logged in using Domain Admin account.
